Pictured cells were grown in formate,unwashed, pictured at an OD600 of 247, high gain image setting, 995ms exposure, and WF refers to WITH Fluorogen (NF denotes WITHOUT Fluorogen), the 4 digit number string represents an arbitrarily numbered field of view from the sample, and FITC denotes fluorescent channel (TRANS indicates transmitted light/phase contrast) A. FruA Micrographs Folder contains images of M. maripaludis expressing an N-terminal FAST1 tag on FruA. Formate Grown and Hydrogen Grown folders contain images grouped by biological replicates of log phase cells grown in media in which either formate or hydrogen was the sole electron donor, respectively. B. Fdh SplitFAST Micrographs folder contain images of M. maripaludis expressing c-terminal fusions of NFAST and CFAST. FdhA+FdhB folder contains cells expressing FdhA tagged with NFAST and FdhB tagged with CFAST. FdhA+Mtd folder contains images of cells expressing FdhA tagged with NFAST and Mtd taged with CFAST. The FdhA+FdhB folder is further subdivided into conditions in which this cell type was imaged grown under hydrogen containing media or formate containg media. -------------------------- METHODOLOGICAL INFORMATION -------------------------- 1. Description of methods used for collection/generation of data: Cells were grown to exponential phase in in liquid media containing either hydrogen or formate as specified above and treated with HMBR to a concentration of 10 micromolar in an anaerobic prep. Images were taken with an echo revolve microscope in anoxy. Images are raw tiff files directly from the native microscope software. They are unprocessed. 2. Methods for processing the data: N/A 3. Instrument- or software-specific information needed to interpret the data: Echo Revolve Microscope The image files are 16-bit (to preserve the light intensity information). To display the images properly they should be opened using a program such as ImageJ am open source supported image viewer that can be downloaded from https://imagej.nih.gov/ij/download.html 4.
