Geographic location of data collection (where was data collected?): Sediment cores were collected from Lake Ann MDNR DOW 10001200 and Lake Riley DOW 10000200 Chanhassen, Carver County, Minnesota. Seedbank analysis was conducted in the Minnesota Aquatic Invasive Species Research Center, MAISRC Containment Laboratory (02-335), 1995 Fitch Avenue, Saint Paul, Minnesota, 55108 5. Information about funding sources that supported the collection of the data: Riley Purgatory Bluff Creek Watershed District, Eden Prairie, MN 55317. Additional support and facilities provided by the Minnesota Aquatic Invasive Species Research Center and the Minnesota Agricultural Experiment Station NIFA grant MIN-41-081. 6. Overview of the data (abstract): Sediment cores (10cm diameter) were collected from Lake Ann and Lake Riley at 1m depth from seven locations around each lake. Sediments cores from each lake were pooled and homogenized and placed in trays with controlled lighting and temperature. Germination was determined for low light conditions (low water clarity), higher light conditions (high water clarity) and a treatment with gibberellic acid to induce germination and sprouting (maximum emergence – called maximum germination in the JAPM publication). Trays were examined weekly for newly sprouted plants that were identified to species and enumerated. -------------------------- SHARING/ACCESS INFORMATION -------------------------- 1. Relationship between files: -------------------------- METHODOLOGICAL INFORMATION -------------------------- 1. Description of methods used for collection/generation of data: Sediment cores (10cm diameter) were collected from Lake Ann and Lake Riley at 1m depth from seven locations around each lake. Sediments cores from each lake were pooled and homogenized. The sediment from each lake was washed with well water to remove large debris and curlyleaf pondweed turions and then 200 ml of sediment was placed into each of 45 19x19x6 cm trays containing 200ml sterilized sand and covered with 3cm well water. Trays (15 per treatment from each lake) were randomly assigned to one of three germination treatments: high clarity (high light; 650 µmol/s/m2), low clarity (low light; 125 µmol/s/m2), and maximum emergence or germination (light = 800 µmol/s/m2 and gibberellic acid was added to a concentration of 0.3 mM to induce germination). Water temperature was maintained between 21-23 ˚C with a 15h light, 9h dark photoperiod. Water was added as needed to maintain 3cm depth covering sediments. Trays were examined weekly for newly sprouted plants. Sprouts were identified to species, removed from the trays and enumerated. The experiment ended after 16 weeks at which time a subset of 5 trays from each treatment was examined for any remaining unsprouted seeds or spores (for Chara). Additional details in Dunne and Newman 2019. 2. Methods for processing the data: Methods are provided above. Means and 2SEs were calculated in excel along with cumulative estimates. 3. Instrument- or software-specific information needed to interpret the data: none 4. Standards and calibration information, if appropriate: none 5. Environmental/experimental conditions: Water temperature was maintained between 21-23 ˚C with a 15h light, 9h dark photoperiod. Water was added as needed to maintain 3cm depth covering sediments. See above for light levels assigned to each treatment. 6. People involved with sample collection, processing, analysis and/or submission: Samples were collected by Melanie Dunne and Raymond Newman.
