Filename: Xylem_anatomy Short description: This file contains data on the timing when xylogenesis begins and and also on average vessel area. B. Filename: Reproductive_anatomy Short description: This file contains data on bud and flower carbon content, water content and growth 2. Relationship between files: All of these files are data collected during a three year project at the Arnold Arboretum investigating seasonal changes in phenology, organ growth and vascularture. 3. Additional related data collected that was not included in the current data package: Yes, there is a second dataset available: Savage, Jessica A. (2019). Retrieved from the Data Repository for the University of Minnesota, https://doi.org/10.13020/5dyn-m363. 4. Are there multiple versions of the dataset? yes/no No -------------------------- METHODOLOGICAL INFORMATION -------------------------- 1. Description of methods used for collection/generation of data: Date of first vessels (start of xylogenesis) was determined by sectioning branches directly under floral/leaf buds with a sliding microtome and staining them with cresyl violet acetate (0.16% w/v). New vessels appear more pink and less blue than mature vessels. See publication for more details. Vessel diameter was determined by sectioning stem samples with a sliding microtome and staining with safranin O. The area of a minimum of 100 vessels were measured in each sample. See publication for more details. Bud and flower mass was measured immediately after collection and then after drying. This allowed us to calculate the average water mass (g) and the dry mass(g) for the tissue. See publication for more details. Flower %N and %C was determined by freeze-drying and grinding flowers and sending them to the Yale Isotope facility for analysis. 2. Methods for processing the data: NA 3.
