Filename: 17SLR_Muskellunge_data_genotypes Short description: Sample data and genotypes for 258 muskellunge, including 60 with acoustic tags, from the St. Louis River estuary of Lake Superior -------------------------- METHODOLOGICAL INFORMATION -------------------------- Muskellunge (Esox masquinongy) were captured in trap nets by the MN and WI Departments of Natural Resources during spawning season in the spring of 2017. Fin clips or scales were collected for genetic analysis. Individuals were genotyped at 13 microsatellite loci as described in Miller et al. 2012; Transactions of the American Fisheries Society, 141:1411-1423. PCR products were submitted to the University of Minnesota Genomics Center for fragment analysis on an ABI 3730xl capillary electrophoresis platform. Allele sizes (in base pairs) were scored by L Miller and E Schaeffer using Genemapper software with the LIZ500 size standard. Based on the genotype data, ancestry of each individual was estimated using the software STRUCTURE (Pritchard et al., 2000; Genetics 155:945–959) with default settings at K=2 and runs of 50,000 followed by 200,000 repetitions. Individuals were then classified into ancestral groups based on their estimated Minnesota strain ancestry: >0.875 = Minnesota strain, <0.125 = Wisconsin strain, intermediate values = strain hybrid. ----------------------------------------- DATA-SPECIFIC INFORMATION FOR: 17SLR_Muskellunge_data_genotypes ----------------------------------------- 1.
